Please note: A correction is needed for the concentration of dNTPs in the multiplex paper. In the paper we report that dNTP concentration is 0.5 mM for each dNTP in the final reaction. The concentration should be 0.3 mM for each dNTP in the final reaction. We apologize for any inconvenience.
To more efficiently collect genotype data for canine genomics projects we have developed multiplex sets for 155 canine microsatellites.
172 markers were originally termed Minimal Screening Set 1 (MSS1) and published as the first set of markers to use in canine linkage studies: Richman M., Mellersh C.S., et al. (2001) Characterization of a minimal screening set of 172 microsatellite markers for genome-wide screens of the canine genome. Journal of Biochemical and Biophysical Methods 47, 137-149.
We've multiplexed 155 of these markers into 48 sets of 2 to 5 markers, and the remaining 17 markers are amplified individually. Amplification of the mutliplex sets is achieved using a single PCR mix and thermocycling program, varying only primer concentrations.

A: tetra-repeat FH2096 labeled with 6FAM, allele sizes of 95 bp and 103 bp.
B: tetra-repeat FH2548 labeled with HEX, allele sizes of 179 bp and 187 bp.
C: tetra-repeat FH2079 labeled with TET, allele size of 275 bp.
D: tetra-repeat FH2985 labeled with HEX, allele sizes of 310 bp and 318 bp.
